Click the Pause button on a connected controller that is not the current active controller. This will setup the controller automatically when the second instance launches.
Wait a few seconds (around 30 if you're not playing with loads of mods), and the second instance should open.
Limitations
As of 0.3.0, only 2 player splitscreen is supported.
As of 0.4.0, 4 player splitscreen is supported!
